1. The purpose of.' thus subproject 47 is to provide funds to continue
a research program being carried out by Carl C. Pfeiffer, MO.
2. The nature of the investigations to be carried out under the
direction of Dr, Pfeiffer encompasses pharmacological and clinical testing
in animals and human volunteers of chemicals and biochemicals which alter
behavior. Certain phases of other problems of interest to TSD will be -
investigated.
3. Dr. Pfeiffer is located at the New Jersey Neuro-Psychiatric
Institute in the Bureau of Research in Neurology^ind^Psychiatry, Princeton,

4. The funds for this pr'ojecfwill be transferred using the

INTRODUCTI ON:
Our research efforts have received local attention and approbation
during the past year so that we now have excellenl^rapport with the staffs
of both the Institute and the *Bordentown -Reformatory . We now have 10
volunteers per week from the latter, and we have a Clinical Investigative
Unit which will house 80 schizophrenics (present census 45 patients).
Current Status of the Pharmacology Unit
The laboratory is still undergoing minor construction, but material
progress has been made in the past year. The section for animal screening
and conditioning is complete. The sections for animal neuropharmacology
and clinical pharmacology have been constructed and put into operation,
andan electronic workshop has been set up. All these units are actively
at work in human and animal experiments as outlined below. The facilities

are already in partial use. Still in varying stages of construction are
the biochemical pharmacology and medicinal chemistry units.
Personnel : Since the last report, the section has been joined by Drs.

as planned. It is anticipated that two to three
more investigators at the .doctoral level will be needed, probably one

more clinical pharmacologist, a medicinal chemist, and a biochemical
pharmacologist. Two engineers have been employed for design and con-
struction of new equipment and six lab assistants are now fully trained
to their various jobs.
Human s ubjects : Liaison with UeLw ^Jersey-State Reformatory — Bordentown
has been excellent, and a full complement of volunteers from that insti-
tution has been freely available. This group may be a better group for
this Kind of research, since they may be closer to the normal population
than were the Atlanta prisoners, the latter being recidivists and older
than the Box n dentown' group. In addition, other human volunteers have
been recruited, including female subjects, so that the population of
«
"normal" subjects available is now much more nearly representative of
the population at large. In addition, it has been possible already to
begin explorations with the psychotic patients from the experimental
ward, and some interesting results are already forthcoming (see below).
Equipment: Since the last report, the section has obtained a stereo-
taxic apparatus, a Grass Model III EEG, and a Sanborn 2000 magnetic
tape Data Recorder. In addition, we have ‘constructed two new integrators
for quantitative EEG; these are modeled after the Drohocki Integrator’;
but due to some circuit changes are more linear’, and have the advantage
of using all USA -made parts, so that maintenance is simplified. Two

integration of four leads, one from each lobe of a hemicranium. Also
under construction is a quantitative spectral analyzer for use with the
tape recorder in analyzing scalp EEG of humans as well as electrical
measurements from implanted electrodes in cortex and deeper structures
in brains of animals. Data will be entered on punch cards and processed
by a digital computer. Target date for beginning calibration and stan-
dardization is June 1, 1962. These new approaches allow determination
with great accuracy of effects of various classes of compounds and the
interactions of such agents.
A Barber Colman gas chroma tograph has been installed and a technician
is being trained to operate it. This makes possible correlative studies
of changes in brain constituents, changes in behavioral state, and electro
physiological data. This method is far more sensitive than previous
techniques, and allows accurate measurements to be made from much smaller
brain fractions. A Beckman pH meter a nd micr obu rette have also been
obtained for study of cholinesterase activity, since it appears that
psychotomimetic effects frequently if not always involve atropine-like
drug action (Pfeiffer, C.C., et al. Neurology, 9: 249-250, 1959)
Recent and current projects : These new techniques, together with the
unique availability of human subjects both normal and abnormal, make
possible a wide range of investigations on any class of compounds with
action on the central nervous system. These can begin at the most basic
level and move progressively to a level of sophistication beyond that of
almost any other setting. Some idea of the directions currently being
taken are indicated in recent and current work. Enclosed are -reprints of
publications during the past year. The following are also accepted for
pubiication and. will appear in the next few months:

tryptamine, 4-methyl -a -methyl tryptamine, 6 -hydroxy-N,N-di methyl tryptamine
(The di-methyl analogue of Szara's compound), px'olyltryptamine, psylocin,
and acetyl tryptamine. Results will be reported ^as^T:hey accrue. The
central actions of ^-aminobutyric acid are being indirectly explored by
investigating y-butyrolaetone and y-hydroxybutyric acid. Unlike GABA,
these compounds readily cross the blood-brain barrier, and the:latter is
a potent hypnotic agent and about as active and positive as is chloral hydrate.

Research Proposals
Tr» the light of the above, the following are planned for the coming year:
1 . Screening of tryptamine compounds in animals and man for hallucino-
genic action. - •;
2. Completion of the study of the use of riboflavin as a trace chemical
to prove the ingestion of medication in a clinical setting.
3. Further studies on the use of phenoxybenzamine and other adrenergic
blocking compounds as antidotes to LSD-25.
4. Further refinement of the use of quantitative brain wave studies to
diagnose schizophrenia and to determine the action of hallucinogens.
5. Selection of one or more of a series of stimulant diamines for chronic
toxicity studies in animals so that they can be tested in normal human
volunteers for possible stimulant action.
6. A search in brain extracts of rabbits treated with Tofranil (iinipra-
mine) to determine if any stimulant amine accumulates as a result of
degradation of the drug molecule in the body.
7. Determination of the anti-cholinesterase and aminoxidase inhibitory
effect of the stimulant diamines.
8. Assay of acet>fcholine content of various portions of the brain after
prolonged administration of deanol.
9. Tire assay in man of an intravenous analgesic solution composed of
morphine and choline or morphine and carnitine wherein the pharma-
cologically neutral quaternary compound prevents the engagement of
peripheral receptors by the morphine.

It Is proposed to conduct screening and range finding tests on volunteers
with chemical compounds of known behavioral activity.: Studies will be
conducted on derivatives of selected biologically active compounds to deter-
mine their action on the central nervous system of man.
In addition to these Investigations, the following work will be undertaken:
1. Pharmacological testing will be conducted on animals
utilizing candidate hallucinogenic chemicals,
2. Studies will be conducted on compounds of relallvly
simple configuration to assess their ability to tnhlbit trained
behavior In animals utilizing the condition avoidance responses.

1. The purpose of this subproject 47 Is to provide funds to continue
a research program being carried out by Carl C, Pfeiffer, MD.
2. The nature of the investigations to be carried out under the
direction of Dr. Pfeiffer encompasses pharmacological and clinical
testing in animals and human volunteers of chemicals and biochemicals
which alter behavior. Certain phases of other problems of interest to

Institute in the Bureau of Research in Neurology and Psychiatry, Princeton,

Proposals A, 8, and C are a continuation of th e/kork now in progress which
is familiar to the officers of The from our annual and semi-
annual reports. Under C th^'source of human volunteers wi 1 1 be the 100 reformatory
inmates (ages 18 to 28) whobre stationed on the grounds of the Institute. If 16
volunteers, aged 21 to 28,- are not obtained frcm this group for a weekly session
of drug testing, we will have one session of eight subjects at the institute and
one session at the New "-'Jersey' State Prison in Trenton. ‘^Commissioner 8rafnberg has
given his permission.
Under £ we now have perfected and plan to use at the New Jersey Laboratory a
quantitative integrator for drug studies in both man and animals.
Adult unanesthetized, trained animals are used. Under anesthesia small gold-
plated wood screws are used as dural electrodes. These are lightly engaged in the
calvarium and held firmly in place with methacrylate with the permanent lead .
attached. The recording leads are bi^pbiar from the left and right frontoparietal
lobes of the brain. The animals are in a sound attenuating box and sit quietly for
the entire 1 to 2 hour session after a few training periods.
A CrohocW integrator is attached to one lead. This device rectifies the brain
waves and by a chopping process, counts the area under the rectified curve. A .
signal is made at a pre-set degree of sensitivity. The number of signals per unit
time is a measure of the degree of electrical activity or ."electrogenesis. 11 The
number of pulses or signals is directly correlatable^wi thT the area under the curve.
The constancy of the electrogenes is with elapsed time can be calculated and treated
by the usual statistical methods. In this type of analysis, periods as long as
five minutes can be used as a baseline or control and thus transient artefacts are
eliminated. The action of drugs is usual ly measured at ] -5 minute intervals after
intravenous injection and the drug action is expressed as change in percentage
electrogenesis from the control preinjection period. Arousal or stimulant drug
effects are recorded as a decrease while sleep (sine yvaves) produce an increase in
electrogenesis.
E. Annual conference on neurobiology of psychoses. Most previous conferences
of this type have been poorly planned, and the group has been too large and unwieldy.
Money spent on verbatim recording and publishing is wasted, end this' process further
inhibits the participants.
The participants will be limited to 12 or 15» The record wiil.be in the form
of the individual’s own notes and the notes of a scienti fical Jy trained secretary.
The summarizing secretary will change with each three-hour session. In a two-day
conference we would have five sessions of three hours each. t

MEMORANDUM FOR: THE RECORD
SUBJECT . : MKULTRA, Subproject 47
1. The. ’purpose of this subproject is to provide funds for
the continuation of the program being carried out at Emory Univer-
sity under Carl C. Pfeiffer, M.D. The scope of this work basically
covers the develo pment Tod testing of new c hemical compounds which
are hallucinogenic or otherwise affect the central nervous system of
hurnans. Certain phasfes of other problems of interest to TSS/CD will
be investigated.
2. The nature of the investig ations to be carried out under the
direction of Dr. Pfeiffer encompasses the synthesis, pharmacology
and pertinent biochemistry of potential hallucinogenic chemical com-
pounds. Testing and evaluation will be conducted in normal human
volunteers.
3. The funds for this project will be transferred using the

of the subjects, such as alertness, sleep and arousal from sleep.

A careful study of SAR of acetylcholine Ach and atropine-like compounds Indi-
cates that the naturally occurring Ach- like compound should have an asymmetric carbon
atom in the acid moiety. The evidence for the existence of Ach inbody fluids Is
tenuous since identification has been made only by Reineche precipitation which
method only tests for quaternary nitrogen containing compounds. A few of the optically
active acids which might be esterified with choline are lactic, pantothenic and as-
corbic .acid;. These analogues of Ach are therefore in process of being synthesized
to determine their pharmacodynamic effect.

chemical systems^ in vitro . Based on the information available at the pre-
sent tine, the systems listed below are being considered for the first se-
ries: of evaluative experiments. While we realize that their causal* connec-
tion wi"h halucinatory disturbances has not been conclusively established,
we feel that they constitute a suitable starting point. In each instance,
known psychotomimetic agents will be evaluated elong with our compounds,

LSD was found to be a powerful inhibitor of pseudccholinesterase in hirzan
■serum and brain. Pseudocholinestere.se activity of rat, guinea-pig, rabbit,
. chicken and nonkey brain is much less sensitive to inhibition bjT LSD than
is the corresponding enzyme in human brain. ^Pseudocholinesterase was in-
hibited more powerfullylbylLSD than by other ergot alkaloids. (R. H. S.

Monoamine oxidase is considered to be a major catalyst of serotonin metabo-
lism in tissues. Iproniazid is reported to inhibit monoamine oxidase in tho
brain, but unable to penetrate to the site of r.onoenine oxidase in intact
cells of other tissues. (H. Vcisrbach, D. F. BogdaneM, B, Redfield end
S. Udenfriend, Fed. Proc., 16: ^*5, )- Relationships be tween monoamine
oxidase end diamine oxidase were reported. There is indication that the af-
finities of the two enzymes are overlaping; however, sor.e of their characte-
ristics have been found to retain their specificities. Mescaline (5,4,5-tri-
nethoxyphenethylRnine) ar.d tyranine were reported to be among the substrates
of diamine oxidase, while some JS-arsinoethyl imidazoles were found to be sub-

hl^The purpose of this project is to provide funds to
continue the research program conducted by Dr. Carl Pfeiffer

viding answers to specific questions of direct interest to the
agency. In addition, those areas of basic knowledge required

The purpose of this memorandum of agreement Is to set forth the
mutual administrative responsibilities of the principal research
investigator and the sponsor. Therefore, it is mutually agreed that,

six month intervals, an informal statement of expenditures of monies
advanced for research activities. It is requested that the statement

Specifically we need for research purposes
in me^al^se^e aEecVrian DK~2 ratio recording Spectrophotometer.
This would be used for the following purposes;
1) Routine assay ^for purity in those compounds synthesized by
or those compounds which he isolates from
natural sources. .
2) Use for monitoring rapid chemical __ reactions in biological
systems,
a) The combination of convulsant acridone with deoxyribo-
nucleic acid or ribonucleic acid.
b) The combination of pyridoxal phosphate with thiocarbo-
hydrazide,
c) The modification of epinephrine by schisophrenic serum
and cerebrospinal fluid. Heath reports a marled difference
in light absorption with time that is characteristic of
schisophrenic serum.
The cost of this basic unit is $7,ll7£.00, but with the vaver
length hold attachment and sales tax the total cost would be about
$8,000.00.

• *
1. The purpose of this addendum is to set forth the mutual
administrative responsibilities of the contractor and sponsor*
2. The above subproject represent the research program of
Dr. Carl Pfeiffer located at Emory University covering the period
of 1 July 1955 through 30 June 1956.
3- The principal In vest igator shall submit to the sponsor,
at six month intervals, an informal statement of expenditures of
monies advanced for research activities. It is requested that
the statement be submitted in essentially the following fora:
a. Personal Compensation (Salaries, Subject Fees),
b. Equip ment ,
c. Suppli e s and Services,
d. Travel,
e . Overhead,
f. Contingencies •
Also, requested Emory University to submit
to them a surer-ary' accounting of monies received from the Fund. They
also requested the return of any unex pended funds received under
a grant from the Fund. L
h. Title to any p ermanent equipment jau chased by funds
granted the University shall be retained by the University in
lieu of higher overhead rates. ‘
5* It vas mutually- agreed -that documentation and accounting
for travel expenses which are reimbursable by the University
shall conform with the accepted practices of that Institution.
6. It vas agreed that technical reports reflecting the
progress of the research program shall be submitted at mutually
acceptable intervals.

MEMORANDUM FOR: THE RECORD
SUBJECT
: Project KKULTRA, Subproject 4?
1. This purpose of this project is to provide funds for the
study of drugs affecting the central nervous system, and to screen
and evaluate pharmacologically an y other com pounds which may be
of interest to J'SS/Chemical Division. ;
2. The attached proposal indicates the extent of the investi-
gations that will be carried out in these facilities available at
Emory University and provides for close cooperation with the work
j ^twmbo transferred using the
usual manner. ^ '
4. The cost of this program for a period of one year begin-
ning 1 July 1955 will be $ 43.803. 1 0 to which must be added four •
percent m fee to making the total amount
$45,555.22. At the present time $15,000.00 is being committed,
$14,400 for the project and $600.00 fee to the r>
The balance of the total will be committed at a later date,
